That's not going to work. Assuming both of your turtles are 4" each or larger because you know the sex of them, a 40 gal would only fit one 4" turtle. Splitting that up gives you 20 gallons per turtle and thats simply too small.
Your looking into a MUCH larger tank, or two seperate large tanks.
That said, they do make seperators for that size tank and you can find them online or in a petsmart or something.
