Don't forget, the EP is a tad bit larger, maybe half an inch to an inch, so I think it'd do fine telling the RES to go somewhere else.

I will get it tested before every putting it in there.

Also, I'm just worried about any chance of them trying to mate...that would be odd, a Red ear painted? Would that even work? I assume it'd be like cross breeding dogs (I really don't want breeding anywho, but if I HAVE to, i will).

By "telling them to go somewhere else" I hope you realize that means fighting. The whole problem is that the females are larger and can easily injure males when they feel angered by their pestering for mating.
I really think that's asking a lot of the female to put up with two sexually mature males.

90 days is really a minimum amount of time considered acceptable for a quarantine period. Many experienced turtle keepers advocate longer.
