I don't think you should be poking your turtle that much or that hard because if there are eggs there, you could break them. If you didn't feel anything hard, then there shouldn't be any eggs. Females display signs of being gravid. This link explains it better then I can.

If she is gravid, then you'll need to fill a fairly large container with sand and soil and put her in it whenever she shows the signs. You'll have to be patient because turtles are picky when it comes to where they want to lay their eggs.

Can I use a tupperware bin how deep should I put it and how to make a nest box in a tank

A tupperware bin is fine.this website says for a larger RES it should be 12 inches deep. So I guess somewhere before that (or 12 inches if you really want) should be fine (I wouldn't go any less then 5-6 inches).
My nesting box is outside of my tank so I don't know how to make one inside. You could ask in the indoor habitat thread how to do it.
